I would use Hylomar on the gasket. I have used this a few times on both my engine and other peoples engines that I have worked on. Put simply, it is good s*&t! The spray on stuff is easier to apply.

I am guessing the Toyota manual would only recommend the use of sealant around the joining between the timing cover and the engine block, this is because of the potential uneveness of this region.

Once again, I would use a smear (note smear, not a whole tube!) around the entire timing cover top lip that touches the head gasket. You might need a slighlty bigger dob around the timing cover/engine join.
